目的通过对肿瘤芯片数据库(Oncomine)和生存分析数据库(Kaplan-Meier Plotter)和蛋白-蛋白相互作用数据库(STRING)进行数据挖掘,探讨干扰素刺激基因15(ISG15)在原发性肝癌中的表达情况及其与患者预后的关系。方法分别对Oncomine数据库、Kaplan-Meier Plotter数据库和STRING数据库进行肝癌中ISG15基因表达的相关研究检索,探讨ISG15基因在肝癌组织和对应正常组织中的表达情况,绘制ISG15基因表达与肝癌患者总体生存时间的生存曲线。构建ISG15相互作用蛋白网络树状图,并对ISG15及其相关蛋白基因进行功能富集分析。结果通过检索Oncomine数据库发现,有5个研究结果显示肝癌组织中ISG15呈高表达(包括细胞系),有2项研究结果显示ISG15在肝癌组织中的表达水平显著高于正常肝组织(P<0.05)。通过对Kaplan-Meier Plotter数据库进行检索分析发现,在病毒性肝炎、饮酒的肝癌患者中,ISG15高表达患者的总体生存时间明显短于ISG15低表达患者(P<0.05)。通过STRING数据库搜索ISG15基因相关蛋白发现,ISG15基因相互作用蛋白有10种;生物过程富集分析结果显示,上述蛋白主要富集在病毒防御和病毒基因组复制的负调控通路。结论ISG15在肝癌组织中高表达,且可能与病毒性肝炎、酒精性肝癌患者的预后密切相关。
ObjectiveThrough data mining on the tumor chip database (Oncomine), survival analysis database (Kaplan-Meier Plotter) and protein-protein interaction database (STRING), to explore the expression of interferon-stimulated gene 15 (ISG15) in primary liver cancer and its relationship with prognosis of patients. MethodsThe related researches on ISG15 expression in liver cancer were searched in the database of Oncomine, Kaplan-Meier Plotter and STRING, respectively, to explore the expression of ISG15 gene in liver cancer tissues and corresponding normal tissues, and to draw the survival curve of ISG15 expression and the overall survival time in liver cancer patients. The Network tree diagram of ISG15 interacting protein was constructed, and the functional enrichment on ISG15 and its related protein genes was analyzed. ResultsBy searching the Oncomine database, it was found that 5 studies showed high expression of ISG15 in liver cancer tissues (including cell lines), and 2 studies showed that the expression level of ISG15 in liver cancer tissues was significantly higher than that in normal liver tissues (P<0.05). According to search and analyze the Kaplan-Meier Plotter database, it was found that among patients with viral hepatitis and alcoholic liver cancer, the overall survival time of patients with high expression of ISG15 was significantly shorter than that of patients with low expression of ISG15 (P<0.05). By searching ISG15 gene-related proteins in the STRING database, it was found that there were 10 interacting proteins of the ISG15 gene. And the results of bioprocess enrichment analysis showed that the above-mentioned proteins were mainly enriched in the negative regulation pathways of virus defense and viral genome replication. ConclusionISG15 is highly expressed in liver cancer tissues, which is closely associated with the prognosis of patients with viral hepatitis and alcoholic liver cancer.
